In this study, we induce a series of low molecular ultra-short dipeptide hydrogelators to explore the self-assembled structure and its physical properties. During the first topic of the research, we design a series of 1,8-Naphthalimide (NI) derivatives through combine with different cyclic amino group. Respectively, NI-FD, 5RNI-FD, 6RNI-FD, 7RNI-FD and 8RNI-FD. We found the nanostructure of odd nμMber ring, 5RNI-FD and 7RNI-FD, is well-defined nanofiber; even nμMer ring, NI-FD, 6RNI-FD, 8RNI-FD appears self-assembled dipeptide nanotube. The most special of this part is that NI-FD and 6RNI-FD is provided with AIE effect, so it represent we probabily successful to synthesize the first self-assembly dipeptide nanotube with AIE property. On the other hand, We observed that 6RNI-FD dramatically increased the fluoresecnce intensity under acidic condition from pH-dependent fluorescence spectrμM,and through MTT assay we also observed 6RNI-FD possess excellent cell-compatibility. For the reason that we use HeLa and PC-3 cell to execute cell imaging with commercial Lysotracker Red DND-99 respectively. Finally, the HeLa and PC-3 cell line all got significant colocalizations 73.05 ± 5.03%and 80.48 ± 3.05% As a result, we recongnized 6RNI-FD has potential on bio-image as a new type of Lysotracker in the future. During the second part of the research, we demonstrated the first self-assembly supramolecular dipeptide hydrogelator with thermal-sensitive property. In this part, we synthesized L-/D-PFB-LF hydrogelator can form hydrogel under physiological environment. Through the increase of the temperature, we observed under 37℃ will promote the gel-forming but room temperature can not. Through MTT assay we found L-/D-PFB-LF possess excellent cell-compatibility toward CTX, 3T3-L1 and hMSCs cell lines illustrate the application on the injectable hydrogel with drug delivery of the L-/D-PFB-LF thermal-sensitive material is very promising.
