Solid state fermentation(SSF) has more active ingredients in the grain matrix and cheaper price than the submerged fermentation. The study was conducted impact of culture conditions on mycelial growth and bioactive ingredients by Ophiocordyceps sinensis in coix lacryma-jobi matrix of solid-state fermentation. The results showed that lactose as a carbon source can be high ergosterol 1.197mg / g, the second carbon source was sucrose can be ergosterol have 1.050mg / g. In the case of different nitrogen source, yeast extract as a nitrogen source can be high ergosterol 0.730mg / g. Solid-liquid ratio of 1: 3, inoculation amount of 10% can be high ergosterol have 1.101mg/g and 0.786mg/g. In this paper, we preferred as the main growth but solid-state fermentation mycelium is not easy to detect so ergosterol content as the main growth indicators that with sucrose as the main carbon source for the rest of the culture conditions at 18℃ and yeast extract as a nitrogen source, inoculation amount of 10%, solid-liquid ratio of 1: 3, the dark for 30 days to get the best ergosterol.
