The metabolic syndrome includes central obesity, hyperglycemia, hypertension and dyslipidemia. Furthermore, many studies have shown that metabolic syndrome is an important risk factor for type 2 diabetes and cardiovascular diseases. Previous study has shown that Chenopodium formosanum Koidz. (CFK) could enhance the glucose uptake of adipocyte. Our previous study also found that short term CFK hush water extract manipulation improved the glucose intolerance in MS rats. Therefore, in this study we investigated the long term effect of the supplementation of CFK husk water extract on metabolic syndrome. The male SD rats are divided into four group:basal group (B), fed with normal chow diet ; control group (C), fed with 60 % fructose diet ; low-dose group (L), fed with 60 % fructose diet plus 0.5 % CFK husk water extract ; high-dose group (H), fed with 60 % fructose diet plus 1 % CFK husk water extract. The study was last for 13 weeks. The blood pressure, fasting plasma glucose, insulin, triglyceride, total cholesterol and abdominal fat mass were measured. The oral glucose tolerance test (OGTT) was also performed in this study. The data shown that the plasma triglyceride, total cholesterol, hepatic triglyceride, systolic blood pressure and abdominal fat mass were significantly increased in group C. Insulin sensitivity of rat in group C was lower than in group B. These data indicated that high fructose diet could cause a cluster of disorders in metabolic syndrome. CFK husk water extract supplementation significantly ameliorated hyperlipidemia, hypertension, insulin resistance (IR) and fat mass accumulation. We also found that supplementation of CFK husk water extract ameliorated IR by enhancing the protein expression content of IRS-1 and GLUT4, and the phosphorylation activity of insulin receptor and Akt.
