Hypertension is the most common risk factor responsible for the progression of cardiovascular and cerebrovascular diseases. Solanum muricatum Ait (pepino) is a popular fruit in Penghu, Taiwan. Residents of Penghu consider that pepino could reduce blood pressure and control diabetes, but the scientific literature associated with anti-hypertensive effects of pepino was less. Thus, we used L-NAME to induce hypertension in ICR mice and investigated the protective effects of pepino aqueous extract. Mice were fed by 1% aqueous pepino extract for ten weeks. Blood and tissues were collected to analyze. Results showed that the intake of pepino extract significantly improved systolic blood pressure and diastolic blood pressure of mice. Furthermore, this extract significantly reduced the protein expression of ACE , aldosterone and arginine vasopressin concentration. The protein expression of ENaC and SGLT1 was also significantly decreased. In addition, this extract significantly increased glutathione content, catalase and glutathione peroxidase activities, as well as total antioxidant capacity in kidney. Pro-inflammatory cytokines such as TNF-α, IL-1β, IL-6 and NF-κB protein expression were also lowered.
