Description
Summary:碩士 === 嘉南藥理大學 === 保健營養系 === 104 === Grifola frondosa (GF), a high value medicinal mushroom in China and Japan, is popularly consumed as traditional medicines and health foods for anticancer and enhancing immune functions. In this study, our aims were (i) to examine the anti-diabetic and anti-inflammatory effects of a GF bioactive compound, ergosterol peroxide (EPO) in palmitic acid (PA)-induced murine C2C12 skeletal muscle cells, and (ii) examine the effects of EPO on insulin, Akt/PI3K, MAPKs and GLUT-4 signaling pathways in PA-induced cells. At 1 to 5 µM significantly up-regulated the glucose absorption and increased the cell growth. 5 µM EPO significantly up-regulated the glucose absorption and down-regulated ROS formation in PA-induced cells. Results on EPO is effect on PA-stimulated cell function revealed that cell viability after PA-stimulated reduced to 80% while EPO protected the cells to 100%. Co-treatment of cells with PA plus 5 µM EPO significantly up-regulated the expression of p-IRS-1, PI3K, p-Akt, and GLUT-4 proteins and down-regulated the expression the activation and phosphorylation of JNK and p38 proteins. These results suggest that EPO may play a potential role in the anti-diabetic and anti-inflammatory effects through up-regulation of glucose up-take, insulin, Akt/PI3K, MAPKs and GLUT-4 signaling pathways and inhibiting the production of ROS.
