The traditional medicinal mushroom, water extract of Ganoderma lucidum mycelium (WEGL), reduces body weight, inflammation, insulin resistance, and modulating the gut microbiota in mice fed with high-fat diet (HFD), the beneficial effects not only displaying in WEGL but also in high molecular (>300kDa) polysaccharides of G. lucidium (G1). However, the interaction between high molecular weight polysaccharide (>300kDa) isolated from G. lucidium (G1) and gut microbiota to reduce obesity is still unclear. Thus, we anaerobically culture feces from HFD-fed mice and chow-fed mice with high molecular weight polysaccharides (G1), and nine days later we separated bacteria and supernatant by high speeds centrifuge. The bacteria and supernatant are intra-gastric gavage everyday to HFD-fed mice. We show that bacteria, which are isolated from anaerobic culture HFD-fed mice feces and G1, have ability to reduce obesity, ameliorate inflammation cytokine expression and insulin resistance effect in HFD-fed mice.
