Department of Biotechnology and Pharmaceutical Yuanpei University of Medical Technology Abstract Many studies have pointed out that tea polyphenols has radio-protecting effect, especially catechins (ex. epigallocatechin gallate) in green tea. However, less research focused on polymeric polyphenols in black tea. The radio-proetcting activities of large-leafed tea on the protecting skin cell from UVB-radiation were investigated in this study. This research examined green tea and black tea (TTES No.12 , 18) extracts which polyphenols composition, antioxidant activity, tyrosinase inhibitory activity and molecular mechanism. The ability to protect the skin cells (CCD-966Sk and HaCaT) from UVB radiation was also investigated. The inhibitory effect of teas on the melanin production in UVB-induced melanoma cells also is investigated. Results showed that total polyphenols, catechins and antioxidant activity of TTES No.18 green tea extract higher than black tea. However, the protective effects of skin fibroblasts and keratinocytes from UVB radiation were higher in blac tea. The HPLC analysis results showed that radio-protecting of black tea come from polymeried polyphenols, such as theaflavins and thearubigins, which exhibit anti-UVB activity through signaling transduction, not exactly free radical scavenging effect of polyphenols.
