Summary:碩士 === 國立陽明大學 === 生物醫學工程學系 === 103 === Over healing response that happens in the vascular wall after injury is a main cause of restenosis, which restricts the success rate of those vascular interventions and may often resulting in limb loss or even death. Therefore, In this study, we developed a system which used non-viral gene carrier to induce selectivity gene expression. This hybrid gene we used can regulate vascular smooth muscle cells growth specificity and be applied to restenosis treatment. We designed the gene vehicles which contain blue fluorescent nano clusters and amphiphilic chitosan. As a non-virus gene delivery system which accompanies with fluorescence imaging functionality, the gene complexes can protect our plasmid gene from degraded by some intracellular enzyme. Complementary gene responded to restrain vascular smooth muscle cells proliferation and migration; on the other hand, the endothelial cells do not have the same effect. In addition, using therapeutic ultrasound can induce endothelial cells proliferation and migration which may improve the efficiency of selectivity treatment between smooth muscle cells and endothelial cells. We expect that the approach we designed may use in Arteries therapy in the future.
