We design the irradiation device to do this research. The beam of blue LED irradiated from the bottom of dish, so the cell were exposed directly without the power decay by medium. Furthermore, there is a mask made by black double-side tape on the irradiation plate. If change the mask with different design at different time point, we could get the results of different time point on the same dish without the change of the cell conditions and use less number of cells. After irradiation, cell viability, apoptosis and oxidative stress were detected by fluorescent staining. The results show that after blue LED irradiation, the oxidative stress was detected in all kind of cells. Nevertheless, the results of cellular damage and apoptosis at these cells were different. Thus, according the results of 6-hour irradiation, distinguish these cells into two types: one is easily influenced by blue LED irradiation, including A375, MNT-1 and NIH/3T3; the other is not, including A-431, HSC-3 and HepG2. The research confirms that the oxidative stress induced by blue LED leads A375, MNT-1 and NIH/3T3 apoptosis.
