In recent year, there are many treatment for degenerative arthritis in the clinical, but these methods have many limitations and disadvantages of the used. Therefore, medical research utilized stem cells or precursor cells to repair osteoarthritis disease, but there are no consensuses in the present years. In this study we comparison bone mesenchymal stem cells and pre-chondrocyte-like cells to evaluate which one is the better effect to mitigate osteoarthritis symptom. In this study, using 100 μM hydrogen peroxide to induce chondrocytes cause inflammatory react, that chondrocytes become to OA-like. Used of Chemiluminescent and gene expression to evaluated its degree of inflammation. Next, induced bone marrow-derived mesenchymal stem cells differentiate to pre-chondrocyte-like cells by TGF-β1. Finally, co-culture with pre-chondrocyte-like cells and OA-like chondrocytes, after co-culture three days, analyzing gene expression to evaluate whether OA-like chondrocytes are convert function back to normal type. In this study, First, Chemiluminescent showed that induced by 100 μM hydrogen peroxide accumulated higher free radical than normal type chondrocytes, and analysis by gene expression showed that pro-inflammatory factor IL-1β, TNF-α has higher performance than normal type chondrocytes. Then, BMSCs induced differentiation by TGF-β1 about 7 to 10 days and we named it pre-chondrocyte-like cell. Finally, co-culture with OA-like chondrocytes by three different methods, BMSCs, Pre-chondrocyte-like cell and none respectively. After three day co-culture experiment and analysis by gene expression. All of the gene expression factor IL-1β, TNF-α, MMP-1, MMP-13, SOX 9, COL 2, ACAN and COL 1, respectively. The results presented that either BMSCs co-culture with OA-like chondrocytes or OA-like chondrocytes have better effect than pre-chondrocyte-like cells. In particular, in BMSCs co-culture with OA-like chondrocytes group indicated that the best treatment with OA-like chondrocytes. In conclusion, according gene expression presents that BMSCs have great effect to mitigate osteoarthritis compared with pre-chondrocyte-like cells. BMSCs are much useful for OA-like chondrocytes convert function back to normal.
