Summary: | 碩士 === 國立臺北科技大學 === 有機高分子研究所 === 103 === It was well-known that the Super Paramagnetism Iron Oxide Nano Particles (SPIONPs) with size less than 30nm are capable of use as a contrast agent of T2 image of MRI. Besides, studies indicated that hyaluronic acid can bind to CD44 receptor on cancer cell surface. This property makes it possible to act as a drug delivery system. However, there are few work emphasizing on the targeting ability of HA. Therefore, we develop HA modified SPIONPs (HA- SPIONPs) and aim to elucidate the abilities of this material on cancer cell targeting and MRI contrast agent.
We first used FITC-conjugated HA to confirm that the HA can attach to the target cancer cells with fluorescence microscope. The chemical and physical properties of HA-encapsulated SPIONPs (HA-SPIONPs) were characterized by DLS, TGA, XRD, SQUID, Zeta potential, TEM and viscometer measurements. Cytotoxicity were carried out to test the biocompatibility and cancer cell targeting ability.
The chemical and physical properties test indicated that the prepared HA-SPIONPs exhibited good dispersion, highly stability and superparamagnetism. The in vitro assay showed that the HA-SPIONPs has no cytotoxicity and exhibited good cancer cell targeting and contrast ability.
Our results showed that HA-SPIONPs have the potential to act as a novel contrast agent for cancer diagnosis on MRI.
