Summary: | 碩士 === 實踐大學 === 食品營養與保健生技學系碩士班 === 103 === Many studies pointed out that hyperglycemia associated with lots of diseases with understanding the inter-relationships between hyperglycemia and related diseases can be more help to prevent disease. Propolis (PPLs) has been indicated the ability of antitumor, antioxidant, anti-inflammatory, and immunomodulatory. The purpose of this study was to investigate the effects of hypoglycemic activities of PPLs in high fat diet fed-Streptozotocin-induced diabetes hyperglycemia rat. Six groups of 10 male Sprague-dawley rats were fed for eight weeks with ( 1 ) control diet, ( 2 ) HF / HFr + STZ with thiazolidinedione ( TZD ), ( 3 ) HF / HFr + STZ ( DM ), ( 4 ) HF / HFr + STZ with 183.9 mg/kg PPLs, ( 5 ) HF / HFr + STZ with 367.8 mg/kg PPLs, ( 6 ) HF / HFr + STZ with 919.5 mg/kg PPLs. Body weight, food intake, water intake, fasting blood glucose ( FBG ), fasting blood insulin ( FBI ), oral glucose tolerance test ( OGTT ), pancreas β cell mass, blood biochemical parameters, antioxidant enzyme activity, proinflammatory cytokines and the expression of related gene in liver and adipocyte were determined. The results show that PPLs supplementation significantly attenuated the increase blood glucose and insulin. Also decrease HOMA-IR, the area under the blood glucose curve in OGTT, total cholesterol, triglyceride, TBARS, proinflammatory cytokines (TNF-α, IL-1β, and IL-6), and the expression of fatty acid synthesis related genes (SREBP-1c, ChREBP, PPAR-γ and FAS) in treatment group. In contrast, the mRNA levels of ppar-α were higher in PPLs treated groups than DM group. Also the expression of insulin sensitivity-related gene ( Leptin, adiponectin ) and pancreas β cell mass were higher in PPLs treated groups than DM group. These datas suggest that PPLs used in this study may effectively improve the diabetes hyperglycemia rats inflammatory conditions, oxidative stress, energy metabolism genes and decrease the damage of pancreas β cell, therefore propolis has the ability to lower blood glucose in hyperglycemic diabetic rats.
