The aim of this study was to evaluate anti-obesity effect of Lactobacillus reuteri strain 263 (Lr 263) and try to understand the underlying mechanism by using high fat diet induced obesity animal model Sprague-Dawley rats. Sprague-Dawley rats were randomly assigned into five groups including one normal diet group (Control; C) and four high-calorie diet-induced obese groups. Each group was given either ddH2O (Obesity; Ob), low dose of Lr 263 (1X-Lr 263, 2.1 × 109 cells / kg BW), the normal dose of Lr 263 ( 2X-Lr 263, 4.2 × 109 cells / kg BW), or high dose of Lr 263 (5X-Lr 263, 10.5 × 109 cells / kg BW) . After 8 weeks, body weight, food intake, food utilization rate, body composition, body fat content and blood serum level, proinflammatory cytokines, the amount of gene expression were measured. The result found that administration Lr 263 can effectively reduce food utilization, decreased body and fat weight, alter body composition profile in obese rat, decreased proinflammatory cytokines ( IL-6、TNF-α), and suppressed fatty acid synthesis-related gene (SREBP-1, FAS) mRNA expression enhanced; fatty acid β-oxidation-related genes (PPAR-α, PPAR-γ) mRNA expression amount. In conclusion, oral Lr 263 may have beneficial anti-obesity effects in obese rats. Thus, these results indicated that Lactobacillus reuteri 263 may have a potential for the treatment of obesity and related disease, and may thought alleviate anti inflation activity, and regulation of metabolic genes expression.
