Due to the aging population is steadily increasing in many countries, more researches investigated on prolong lifespan and reduce age-related diseases. Traditional Chinese medicine often used as a medicinal cuisine in Taiwan, hundreds of herbs have been used to longevity, such as Ganoderma lucidum that rich in polysaccharides. Firstly, the present study investigated the lifespan-prolonging activity of dietary mushroom (Hericium erinaceous, Lemtinula edodes and P. sajor-caju) hot water extracts in w1118 wild type male fly strain. Results revealed that H. erinaceous group could significantly extend the lifespan of flies. As a result of the highly association with aging and free radical, furthermore, we analyzed the H. erinaceous of antioxidant activity, composition and the enzyme of SOD, CAT, Ubi in fly, and explored its underlying mechanism of anti-aging. The H. erinaceous hot water extracts (HEW) through DPPH radical scavenging, reducing powder and TEAC assay confirmed that HEW has antioxidant activity. The composition of HEW shown total polyphenol was 6 μg gallic acid/mg, total polysaccharides was 149.2 μg glucose/mg. Western blot analysis demonstrated that HEW could up-regulation SOD, CAT and down-regulation Ubi in flies. In addition, flies exposed to the superoxide generator and neurotoxic agent paraquat test shown HEW could increase oxidative stress tolerance. It was concluded that H. erinaceous rich in polysaccharides and has antioxidant ability also suggests that H. erinaceous could increase oxidative stress tolerance and promote longevity in flies.
