It has been known ubiquitin is increased in liver cancer. Chrysanthemum (Chrysanthemum morifolium), an edible herbs, is used to decreased irascibility. However, there is no study to provide the evidence that chrysanthemum could block ubiquitin-proteasome pathway to prevent liver cancer. Two abundant compound, luteolin (3',4',5,7-Tetrahydroxyflavone ) and borneol, are in chrysanthemum. We demonstrated that luteolin or combination with borneol inhibited 26S proteasome and E1 activity. Luteolin alone has better inhibition than combination of luteolin and borneol on 26S activity. Combination of luteolin and borneol disturb the 26S proteasome structure. Luteolin had a great impact on the cell viability and 26S activity in HepG-2 cells. Pre-G1 of cell cycle were observed in high concentration of luteolin or combination with borneol-treated HepG-2 cells.
