Metalloproteinases (MMPs) and reactive oxygen species (ROS) are increased by UVB treatment. Baicalein, isolated from the roots of Scutellaria baicalensis, is used for protection of photoaging. Baicalein is not only a ROS scavenger but also an inhibitor of 12-Lipoxygenase, has the ability of anti-inflammatory and anti-oxidant. 12-Lipoxygenase catalyzes arachidonic acid to 12-HETE which is an activator of TRPV1 channel. In this study, we demonstrated that UVB increase MMP-1protein level in calcium-dependent manner but not ROS. We confirmed that 12-HETE increase intracellular calcium via TRPV1 ion channel. Treatment TRPV1 blocker can also cause decreased MMP-1 by UVB irradiated. Baicalein can reduce MMP-1 protein level through inhibit 12-HETE generation . On the other hand, silence TRPV1 channel can reverse affect by UVB irradiated.
