According to previous literature, neural stem cells promote glioma growth, and the evidence of postmenopausal women treated by teriparatide increased the population of hematopoietic stem cells (HSCs). Since HSCs may differentiate into various kinds of blood cells and endovascular cells. We therefore hypothesized that teriparatide might promote glioma growth through HSCs-induced angiogenesis. C6-E2 crimson glioma cells were xenografted into murine brain. Mice underwent teriparatide injection to murine femoral muscle from day 7 to day 12 (total six injections each mice) using the dosage of 20 μg/80 μL in drug-treated group. Mice were sacrificed on day 14. Xenografted brains were harvested for hematoxylin & eosin, immunohistochemical (IHC), and immunofluorescence (IF) staining to quantitate orthotopic glioma size, HSCs and blood endothelial cells between teriparatide group and control group. The average glioma volume was 6.17±5.13 mm3 in teriparatide group, and 2.28± 1.62 mm3 in control group. The CD34 positive IHC staining and CD31 positive IHC staining were greater in teriparatide treatment group than in control group (29.67±17.7 cells vs 14.2±4.41 cells and 19.2±22.3 cells vs 0.16±0.37 cells) .This study demonstrated the C6 orthotopic glioma volume, CD34, and CD31 expressions in teriparatide treatment group are significant greater than control group, suggesting that teriparatide promotes glioma growth through CD34 positive HSCs-induced angiogenesis.
