The standard treatments for oral cancers are surgery, radiotherapy and chemotherapy. The compounds isolated from the soft coral Sinularia flexibilis have been reported anti-cancer effect. This study focuses on the screening of anti-oral cancer compounds isolated from soft coral Sinularia flexibilis. The soft coral extracted with Ethyl acetate and then was concentrated under vacuum to obtain a crude extract (40.5 g). The crude extract was separated on the column chromatography to furnish 11 fractions. Six compounds mainly from 3 fractions were structurally identified by NMR and were tested for the anti-tumor effect with oral cancer SAS cell line by MTT assay. The results showed that there were two compounds (sinularin and dihydrosinularin) with IC50 values at 8.2 and 14.2 μM in SAS cells, respectively. Sinularin and dihydrosinularin could induce G0/G1 cell cycle arrest and increase levels of LC-3 protein, while caused the decrease in Cyclin D expression. Further combination of compounds and chemotherapy drug could inhibit cell proliferation of SAS cells. Taken together, the anticancer effect of sinularin and dihydrosinularin are associated with the cell arrest and autophagy. These results suggest that sinularin and dihydrosinularin from marine compounds derivatives exert as a potential drug for the development of anti-oral cancer agent.
