Hepatocyte growth factor (HGF) is a pleiotrophic cytokine, capable of inducing responses on typeⅡ pneumocytes for growth-prompting activity via interaction with a single receptor, MET, in various species but haven't been reported in swine. The present study aimed to explore the causal role of HGF and MET signaling activation on type II pneumocytes proliferation of PNP in swine. Pneumonic pig cases (n=32) and control pigs without respiratory infections (n=3) collected between 2014 and 2015 from Yunlin-Chiayi-Tainan animal disease diagnosis center carrying out quantitation of HGF expression level in the cells collected from bronchoalveolar lavage fluid (BALF) measured by real-time PCR, immunohistochemistry (IHC) detection of MET and PNP lesion scored to lung lobes. The results revealed that PNP samples are remarkably higher than normal ones in HGF expression level (p<0.05) and MET receptor activation (p<0.01), furthermore, PNP lesion scores represent positive correlations either with HGF expression level (r=0.43) and MET receptor activation (r=0.70). The study demonstrates HGF/MET acting on porcine lung with typeⅡ pneumocytes growth-prompting biologic function just as other species, as well as imply a correlation between PNP lesion severity and the activation of HGF/MET signaling, being one of pathway involved in PNP in swine.
