The induction was achieved by using juvenile leaves as explants incubating on Murashige and Skoog (MS) medium supplemented with 5.37μM α-naphthalene acetic acid (NAA). There was no significant difference in the GGB induction using either proximal or distal half leaves as explants. The inoculation test of homogenized GGB tissues revealed that the single inoculum with 0.05 g fresh tissue per Petri dish was better than those of 0.1, 0.15 and 0.2 g (p < 0.05); and subsequently obtained a production of 121.2 shoots per dish. The addition of 2.5 g L-1 activated charcoal (AC) on culture medium with 0.05 g dish-1 inoculum greatly enhanced the regeneration of juvenile sporophytes (362.0/dish). However, the addition of polyvinylpyrrolidone (PVP) or polyvinylpolypyrrolidone (PVPP) was less effect as compared to AC. The elongated sporophytes (1-1.5 cm in length) were rooted successfully by auxin stimulation where a 6 μM-indolebutyric acid (IBA) treatment achieved the best rooting percentage (80%) and also obtained the greatest root number (4.0/explant). Rooted shoots performed 100% survival rate and maintained greater root number (28.8/explant) through the acclimatization trials when planted in the peat moss rooting medium.
