Infected fishes by megalocytivirus belonging to family Iridoviridae appear organomegaly and cause death, so understand the mechanism of viral infection and finding prevention methods to solve this problem, and also the major studied propose in this thesis. In this thesis, the use of infectious spleen and kidney necrosis virus (ISKNV) for the study, to infect GF-1 cell, in the process to find within this infection that increasing ROS could increase pro-apoptotic genes such as Bax, Bak, and tBid, and reduce anti-apoptotic Bcl-2 and Bcl-xL, thus enhance mitochondrial membrane potential (MMP) loss, and activate caspase family such as caspase-8, caspase-9 and caspase-3, so finally cause cell death. Antioxidants treatment could reduce expression of pro-apoptotic genes, affect expression of anti-apoptotic gene, reduce MMP loss, and inhibit activation of caspase family, finally increase cell viability. On the other hand, they could reduce anti-oxidative enzymes SOD and catalase and inhibit viral replication. In sum, viral infection could induce ROS increasing to regulate expression of pro-apoptotic genes causing cell death, and antioxidants treatment could inhibit this result, regulates the balance of oxidative stress, and reduces viral replication.
