Gonadotropins (GTHs), FSH and LH are the key regulators of gonadal development and reproduction. These hormones are heterodimers, consisting of the noncovalent association of a common α subunit with a unique βsubunit that confers biological specificity to the hormone.However, GTHs sequence between mammals and birds is very different, the effects of commercial mammal-GTHs on birds needs further confirmation. Furthermore, less information regarding the different function of birds FSH and LH is available. In this experiment, wecloned complete sequence of CGα、FSHαand LHβcDNA from chicken and used NCS peptide sequence as a bridge connecting the recombinant hormone α-subunit and β-subunit. The results showed that given rooster(week9) testicular tissue exogenous recombinant FSH and LH, CREM gene expression had significantly increased (p <0.05). It indicated that this approach expressing recombinant hormone with RCAS system can successfully activate FSH/LH pathways. In order to comprehend the regulation of FSH and LH in biological mechanisms on reproductive function of chicken, individual exogenous recombinant hormones were given to subcutaneously regenerate testicles. The results showed that the relative volume of subcutaneously testicles in each groups increased with age. LH treatment group has significant highervolume than control group between week2 to week7after transplantation(p <0.05).Interesting, given exogenous recombinant FSH and LH to rooster before sexual maturation. The result showed that FSH treatment group had significant higher Germ cells number than Control group (p <0.05).These different results suggest that theregulatory mechanism of FSH with LH in different stage or condition plays a different role on control gonadal development and reproduction. 鄭旭辰 2015 學位論文 ; thesis 104 zh-TW
