Summary:碩士 === 大葉大學 === 分子生物科技學系碩士班 === 103 === Lung cancer is one of the most common cancer in the world. Metastasis is the main points making lung cancer patient dead. There are many natural extracts is being researched to inhibit cancer proliferation and mobility. α-Solanine is the natural glycoalkaloid in potato, in recent research, α-Solanine was identified which can inhibited cell proliferation in breast cancer cell, colorectal cancer, liver cancer, and prostate cancer, but it is unclear in lung cancer. We used a highly-metastatic lung adenocarcinoma cell line CL1-5 as a model, treatment CL1-5 cell lines with α-Solanine in dose-dependent manner, we found α-Solanine inhibit lung cancer cell proliferation, colony formation, migration and invasion, even made lung cancer cell lines CL1-5 change the phenotype. Finally we used Microarray to compare with the gene expression of CL1-5 lung cancer cell lines with different dose of α-Solanine, then we used David functional annotation tool to analyze downstream signal pathway. Our result suggested α-Solanine is able to inhibits not only cell proliferation via NFкB pathway but also reduces cell mobility by rearrange the cytoskeleton.
