We focused on the detection of the adeIJK on genomic DNA of A. baumannii by PCR and DNA sequencing then determined the variety of antibiotics susceptibility by treatments of efflux pump inhibitors. Firstly, we detected the adeIJK genes in isolates of 230 multiple-drug resistant A. baumannii by PCR, sequencing and blasting Gene Bank. Secondly, we measured accumulation of H33342 and antibiotics susceptibility by disk diffusion tests of tigecycline, minocycline and colistin measured in 2 reference strains and 4 clinical isolates with and without efflux inhibitors, carbonyl cyanide-m-chlorophenyl hydrazone (CCCP). Results show all A. baumannii isolates having adeIJK gene in genomic DNA, and the accumulation of H33342 were not significantly increased in 4 tested A. baumannii clinical isolates by CCCP treatment. Additionally, the diameter of inhibition zone of antibiotics, tigecycline and colistin, were increased in several A. baumannii isolates by treatment of CCCP. Results indicated that resistant isolate E0035 became intermediate to tigecycline; the intermediate isolates E0526、E0610、E0445、E0634 and E1365 became susceptible to tigecycline, in addition, the intermediate isolates E0634 and E1365 became susceptible to colistin. Our results indicate that the adeIJK is prevalent in genome of A. baumannii isolates, and the efflux pump-based system may play some role in tigecycline resistance in A. baumannii isolates.
