The miRNA plays a critical role in cells, including development, differentiation, apoptosis, metabolism, proliferation, tumorigenesis and metastasis. Previous studies indicate that miRNA could be a biological marker or the predictor of prognosis and drug resistance in cancer. The study of miR-338 in cancer is rare, and there is no report in breast cancer. Therefore, we would like to investigate the role of miR-338 in breast cancer carcinogenesis. In this study, we found that overexpression of miR-338 would supprress cell growth, viability, migrated and invaded abilities. We also found that overexpression of miR-338 would not affect the cell cycle-associated proteins, but affected the EMT-associated proteins and decreased the YY1 oncogene expression, it showed that miR-338 may suppressed breast cancer carcinoma by targeting oncogene. Furthermore, miR-338 decreased the luciferase activity of YY1 3’UTR reporter plasmid, and also decreased YY1 expression both in mRNA and protein levels. Moreover, we used miR-338-3p or -5p inhibitors to study which one plays the critical role in breast cancer, we found that either inhibited miR-338-3p or miR-338-5p could increase migrated ability.
