Oleic acid was selected as an oil phase, polyoxyethylene (20) sorbitan monolaurate (Tween 20) as a surfactant, transcutol as a cosurfactant and water as an immiscible phase of the microemulsions to encapsulated RK. To assess the physical and chemical characterizations, we analyze the stability of the particle size, morphology, viscosityand conductivity of microemulsion. The microemulsions with different surfactant/co-surfactant ratio were prepared. Ex-vivo permeation studies were performed with Franz-type diffusion cells to measure the microemulsions transdermal delivery of RK into porcine ear skin. Afterward, the best transdermal delivery microemulsion was analyzed its cytotoxicity and functions. Melanogenesis inhibition by RK was investigated in vitro in cultivated murine B16-F0 melanoma cells. The results showed that a concentration of50ppmRK (dissolved in DMSO and in ME2) could not reduce the relativemelanincontent, comparing to the positive control arbutin. Free RK or in microemulsion inhibited melanogenesis ineffectively. In this study, we investigated theanti-adipogenic effect of RK on preadipocyte cell line3T3-L1.The RK and ME2-RK were incubated with 3T3-L1 at twodifferent stages:preadipocyte proliferation and after differentiation, toevaluate the potency of inducing adipocyte differentiation and lipid accumulation in mature adipocytes.Compared to the control,25 ppm RK could reduce about 20%mature adipocytes or lipid accumulation.It is concluded that RKreduced adipocyte differentiation and lipogenesis in mature adipocytes. This study demonstrated that microemulsion containing RK is an effective transdermal delivery vehicle, and low cytotoxicity.
