The first part is to prepare hollow Fe/SiO2 composite spheres by using poly(acrylic acid) as a template and TEOS as a precursor for SiO2 hollow spheres, followed by depositing Fe nanoparticles on hollow spheres with a chemical reduction reaction conducting in an emulsion solution. To avoid the oxidation of Fe nanoparticles, the Fe/SiO2 hollow spheres were covered with a thin SiO2 layer. The final diameter of the Fe/SiO2 hollow sphere is 200±20 nm. The second part is to prepare Fe/SiO2 composite spheres instead of hollow spheres with three different concentrations of FeCl3 solution. The SiO2-protected Fe/SiO2 composite spheres had the diameter of 200±20 nm.The methods to characterize the Fe/SiO2 spheres are XRD、EDS and ICP-MS to confirm the structure and composition of Fe nanoparticles. Microstructure was characterized by TEM and the magnetic performance was measured by SQUID. The third part studies the drug release experiment of Fe/SiO2 composite spheres, compared to that for SiO2-only spheres. The last part is to study the biological compatibility by WST-8 cell viability test、Live/Dead staining、TEM ultrathin section, and optical image from cell co-culture with drug carriers. The cell used in biological compatibility is L929 together with SiO2 spheres and Fe/SiO2 composite spheres. The result showed that the cell viability of Fe/SiO2 composite spheres was lower than that of SiO2 spheres. The similar result also showed in live/dead staining image. From the TEM image, it had observed the composite spheres inside the cell because of the cell endocytosis behavior .
