So we can analyze the bacterial structure and the difference by sampling from different environment. In this study, various seawater and sediment samples in the offshore of Kaohsiung Harbor in southern Taiwan and Dongsha island were collected and analyzed for the non-cultivated bacteria in each sample. In addition, various seawater and sediment samples in the offshore of Kaohsiung Harbor were analyzed for the cultivated bacteria. Molecular techniques including PCR, denaturing gradient gel electrophoresis (DGGE), cloning and 16S rDNA sequencing were used to reveal the microbial diversities. The results showed that the bacterial isolated found in the non-cultivated samples of Kaohsiung Harbor were similiar, however, with different proportions. Gammaproteobacteria were predominant species in the cultivated schemes, the next abundant species were Bacilli. The bacterial community diversity are much in the non-cultivated schemes of Kaohsiung Harbor than in the cultivated schemes. In the sediment bacterial community of the both cultivated and non-cultivated are the most in the Bacilli specie. But the Gammaproteobacteria specie is predominant in the seawater bacterial community of the cultivated scheme, and the non-cultivated is the Alphaproteobacteria specie. The non-cultivated bacterial community diversity of seawater of the Kaohsiung and Dongsha island are smiliar. However, the species of Deltaproteobacteria, Fibrobacteres/Acidobacteria group, Lactobacillales, Prochlorales, Verrucomicrobia are only found in the samples of Kaohsiung Harbor, and the species of Epsilonproteobacteria, Anaerolineae, CFB group, Chloroflexi, Thermomicrobia are only existed in the Dongsha Island.
