First scale epithelium was to remove by 0.3 M NaOH, and then 0.3 M HCl was to use to obtain the deash of the fish scale. Each 40 ml tube containing 2 g of fish scale under the same water condition of 0, 0.5, 1 and 2 M concentration of acetic acid solution were extracted. After every 4, 8, 12, 16, 20, 24 hour time point, The Coomassie brilliant blue G-250 was used to determinate the concentration of the collagen for each sample. With 2 M acetic acid solution 100oC water extraction for 20 hours was obtained the maximum collagen concentration of 678 μg / ml. The result showed that the water extraction time and acetic acid concentrations can affect the dissolution amount of collagen in addition, the longer the time, and the higher the concentration acetic acid were effectively macromolecules degraded the collagen into smaller molecules amino acid. Extraction times were from 0 to 24 hours, the highest concentration of extract time was between 16 and 20. After sterilization and freeze-dried process, the different percentage of collagen powder (1%, 2% and 3%) was added to cosmetic formula, including toner, essence, lotion, and cream, and then test their stability and resistance. There were no precipitate collapse, stratification and oil phenomenon in toner and cream, but only the lotion formula with 3% collagen powder presented the started precipitates after placed in room temperature for two weeks. Precipitate started to crystalize after placing three weeks, as well as the 2% one. Resistance test in circulation oven and ultra-low temperature test both showed no sign of precipitate, collapse base, stratification and oil phenomenon. In addition, essence containg 2% and 3% of collagen powder appeared layered micro-collapse after the centrifuge seperation test. The four different concentrations of collagen cosmetic tested with Coomassie brilliant blue G-250 showed that the higher concentration of collagen added in cosmetic was measured also higher. The striped bass fish scale are rich in collagen that can be used as a natural moisturizing cosmetic raw material sources.
