Strains used in this study by the National Kaohsiung Normal University mud nests are isolated from the pool area of NKNU department of biotechnology after Dr. WangHui Liang identified Bacillus amyloliquefaciens A1. In this study, different culture conditions A1 fermentation for spore concentrations and metabolite production in cultured explore optimal conditions. The results showed that the trace metals, 0.001mM FeSO4, 0.45mM ZnSO4, 0.001mM CuSO4 and 0.75mM K2HPO4added on cultured Bacillus amyloliquefaciens A1 obtained spore concentration of 10.30 log CFU / mL, the highest surfactin yield of 2296.19 mg /L, and Iturin A yield of 132.73mg / L. In the maximum of 120 hours after adding 100 mesh type coffee grounds as a solid carrier, allowed to stand at ambient and pH 5.5 for one and two hours, spore concentration was 2.00 log CFU / mL and 1.47 log CFU / mL. Performed with 1% molasses fed batch shake flask culture sugars highest surfactin yield, was 1742.75 mg / L and 1795.63 mg / L; 1% golden sugar to be fed-batch shake flask culture sugars to 48 and After 120 hours Iturin A has the highest yield was 203.11 mg / L and 143.14 mg / L. In the 5-liter fermenter 0.1% coffee grounds as solid vectors, and pH-stat as 2% golden sugar fed fed-batch culture indicators, obtain higher spore concentrations 11.69 CFU / mL; secondary metabolite production part, surfactin and Iturin A higher yield production reached 4179.66 mg / L and 301.47 mg / L.
