Photodynamic therapy (PDT) is the best choice as compared to the chemotherapy for its side effects and treatment using PDT can be repeated. Light (Laser) source plays a pivotal role at suitable light-sensitive wavelengths for photochemical effect generated by light-sensitive substance to generate free radicals responsible for cytotoxicity in cancer cells The treatment depends on the cumulative effect of the light-sensitive material and the effective light dose target site; but phototoxic damage straight to be further explored. Nanomaterials - layered hydroxides (Layered double hydroxides, LDHs) can deliver light sensitive material (PdTCPP) confined to the site of action and this study focuses by reducing side effects therefore advantages to improve concentration and reduce the dose of the drug. Since PdTCPP, a photosensitizer with low stability and low bioavailability; significantly reduce the effectiveness of its photodynamic therapy, which limited its clinical application. Nanocomposites LDH-PdTCPP, size measured by electron microscopy and nano particle size analyzer to at size of 150-200 nm, X-ray diffraction (XRD), Fourier transform infrared spectroscopy confirmed the light sensitive substance PdTCPP intercalation in LDH-bit mode annexed to be the sandwich. TGA confirms the thermal stability of PdTCPP in LDH nanomaterials. PdTCPP at 532 nm Laser treatment can generate singlet oxygen (1O2). LDH-PdTCPP on subcutaneous B16F10 mouse melanoma is cytotoxic (IC50 = 22.6 μg / mL), lactate dehydrogenase test (LDH assay) did not make inferences of LDH-PdTCPP (25 μg / mL) on B16F10 cells to necrosis (Necrosis ) of the path; Comet assay proved LDH-PdTCPP Laser (25 μg / mL)caused DNA damage. In addition, JC-1 dye was used to detect color change ratio metrically for determination of decrease (LDH-PdTCPP Laser 20 s, 25 μg / mL) in mitochondrial membrane potential, thereby releasing Cytochrome C indirectly, which lead in apoptotic cell death.
