Summary: | 碩士 === 國立彰化師範大學 === 生物學系 === 102 === Obesity is one of modern civilized illness. It not only affects our health, but also reduces productivity, improves health care costs. Obesity may result from genetic, diseases and drugs, even living and eating habits. No matter what is the reason, the development of obesity is characterized by increases in the number and size of mature adipocytes in the body produced by differentiation and mitogenesis. Many studies have pointed out that polyphenols have anti-obesity function, and seaweeds contain a lot. Compared with other crops such as grapes, tea and apples also contain a lot of polyphenols, seaweeds have higher productivity and lower costs. In this study, we established a 3T3-L1 preadipocyte culture and differentiation system to screen the inhibiting agents of the differentiation and lipid accumulation using algal extraction. The cells grow in 96-well plate with FBS-DMEM medium. After reaching confluent, add the algal exactions as treatment and its concentrations were 0, 12.5, 25, 50, 100, 200, and 400 μg ml-1 phlorlglucinol, respectively. The algal extraction including three brown seaweeds Sargassum thunbergii, S. polycystum, S. aquifolium, and one red algae, Melanamansia glomerata. Cell viability and lipid accumulation were analyzed by MTT assay and Oil-Red O, respectively. The results showed that in the differentiation stage, seaweeds significantly inhibited the cell viability of cells, but it also showed toxicity to the cell. After differentiation stage, treated with seaweeds extraction can not affect the cell viability and the lipid accumulation. The results suggests that Sargassum and Melanamansia might not be appropriated materials for weight control.
