Summary: | 碩士 === 國立中興大學 === 生命科學系所 === 102 === Leptin is a polypeptide hormone playing an important role in increasing metabolism, reducing appetite and regulating weight. In animals with leptin deficiency such as ob/ob mice, the regulation of energy homeostasis by leptin is impaired and animals exhibit increased appetite and reduced peripheral metabolic rate. The excess of body weight in ob/ob mice can be reduced by repeated injections of recombinant leptin. Injections of leptin into normal mice had similar changes such as weight loss, decreased food intake, and a reduction of body fat. At present, there are two popular varieties of Taiwan country chicken. Red-feathered and black-feathered Taiwan country chicken respectively. The red-feathered Taiwan country chicken has a body weight heavier than black-feathered Taiwan country chicken. The black-feathered Taiwan country chicken’s meat has higher quality than the red-feathered Taiwan country chicken. The smaller body size has slower growth rate, so we want to improve the mechanism. By inhibiting the function of leptin to increase appetite and body weight. Using a fusion protein containing leptin repeats of the potential epitopes, and this antigen will induce autoimmune response to endogenous leptin in chicken. After feeding the antigen, The black-feathered Taiwan country male chicken had no significant difference between treatment and control in growth trait. But in female, there is a significant difference between treatment and control in growth trait. The black-feathered Taiwan country female chicken after feeding an additive was effective in increasing the body weight at 9, 10, 11, 14 and 15 weeks of age.
