The advantages of this eco-plasticizer have cost-effective, low environmental mobility hazards, and different types of products. Although eco-plasticizers are environmental friendly with a number of advantages, the toxic effects are not yet understood. This study is focus on investigating the biological effects in toxicity testing for these commercial eco-plasticizers. Toxicity tests were conducted in cell experiments with skin contact and ingestion exposure pathways. Both the skin (A431 cells) and intestinal epithelial (IEC 6 cells) cell lines were respectively used for submerged and air-liquid interface exposure models. The short-term exposure period as acute toxicity and 24 hours post-exposure culture to toxic plasticizer comparing to soybean oil as a reference toxicant were evaluated with cytotoxic cell viability test. Experimental results show that the cell viability from both the skin and intestinal epithelial cell lines were decreased more than 50% in both submerged and air-liquid interface modes exposed to eco-plasticizer in 500 μ1, 5 minutes. The cytotoxicity of 24 hours post-exposure was even lower than those of short-term exposure. The relative toxicity by comparing eco-plasticizer to soybean oil, the eco-plasticizer has higher toxicity than the soybean oil. The results of toxicity tests showed that the eco-plasticizer in the short-term exposure can cause cell damage both in the skin and intestinal epithelial cell lines. This model system could be further applied for other oil toxicant assay in cell culture system.
