To reduce the cardiovascular disease, several studies in reducing the total plasma cholesterol and preventing the LDL oxidation were conducted among worldwide. Feruloyl oligosaccharide (FAO) was found to exhibit more water solubility and antioxidant activity than free ferulic acid. FAO was also show the significantly effect in LDL oxidation inhibition. In order to evaluate the feasibility of FAO as a prebiotic, screen a potential cholesterol-removal strain of Lactobacillusspp., effect of FAO to the cell growth and the functions of fermented supernatant were examined in this work. To screen a potential cholesterol-removal strain of Lactobacillus.is based on the criteria of its bile salt hydrolase (BSH) activity, cholesterol assimilation ability, cholesterol adsorption ability and relative gastric acid tolerance. L. reuteri was selected from 27 strains of Lactobacillus spp.in this work. L. reuteri was found with 0.57±0.07 U/mg of BSH activity, can remove75.09±1.60% of cholesterol from medium, can remove 45% cholesterol by dead cells from medium and have 105 cfu/mL survivals after incubated under pH 2 medium for 2 hrs with a initial 108 cfu/mL cell concentration. In the time course study, results showed L. reuteri growth was in proportion to the cholesterol depletion. This demonstrated cholesterol could be a nutrient that L. reuteri could be uptaken for growth. Furthermore, FAO was found superior than other carbon sources in stimulating L. reuteri growth. After incubated at different carbon sources for 24 hours, the cells collected from FAO media also show the related higher BSH activity and degree of acid tolerance. And the media after fermented with different carbon sources, media from FAO fermented show higher peroxyl radical scavenging activity and correspond to 0.1mg Trolox.
