Several studies have considered that oxidative stress not only affects the expression of heme oxygenase (HO)-1 and Glutathione S-transferase pi 1 (GSTP1), but also brings out cell apoptosis. Brassica oleracea sprout is accounted as a natural antioxidant, which contains vitamins and other bioactive compounds. The aim of this study is to investigate whether Brassica oleracea sprout extract could effectively assist ARPE-19 cells in resistance to hydrogen peroxide (H2O2)-induced oxidative stress by regulating the expression of HO-1. By MTT assay, Western blotting, and immunocytochemistry, we found that ARPE 19 cells, a human RPE cell line, treated with 1 mg/ml Brassica oleracea sprout extract for 24 hrs can protect the cells from oxidative damage by inducing the expression of HO-1, but not GSTP1, in the cytoplasm. The result of DCFH-DA assay also showed Brassica oleracea sprout extract can significantly inhibit the ROS production in ARPE 19 cells, confirming the anti-oxidative effect of the extract. In addition to the expression of HO-1, the level of advanced glycation end-products (AGEs) was also affected by the Brassica oleracea sprout extract treatment. The results suggested that ARPE 19 cells with appropriate Brassica oleracea sprout extract treatment could induce HO-1 expression in the cytoplasm to protect the cells from oxidative damage.
