The major bioactive constituents of AC are triterpenoids which reported to have various activities including anticancer and anti-inflammatory potential. There are two types comprising ergostane and lanostane of triterpenoids are reported from AC. The quality and content of active compounds in AC are dependent on harvest period, extraction methods and culture conditions. Thus, the quality control of active constituents in AC is of great importance. To evaluate the quality of AC, in this study, we developed an accurate and rapid LC-ESI-MS/MS analytical method for the determination of five bioactive triterpenoids namely dehydroeburicoic acid, 15α-acetyldehydrosulphurenic acid, antcin B, zhankuic acid C, antcin A, antcin C and antcin K. The optimized mass spectrometry operating conditions were as follows: negative ionization mode, scan spectra from m/z 150 to 700, nitrogen flow rate of 5 μl/min, capillary temperature at 400 °C, nebulizer gas pressure 15 arb and voltage 3 kV. An amplitude voltage of 39 - 42% and 30 - 35% was typically used for lanostane and ergostane triterpenoids, respectively, fragmentation in the ion trap auto-MS2 experiments.
