However, certain studies revealed that PAMAM dendrimers impaired mammalian cells. Moreover, in terms of teratogenicity study little information focused on the cytotoxicological study of PAMAM dendrimers on mammalian embryos. Thus, this study is conducted to examine the influence of generation 2 (G2) and generation 3 (G3) of PAMAM dendrimer on mouse blastocyst. To investigate the influence of PAMAM-induced cytotoxicity, cell morphology, apoptosis, proliferation, in vitro development, reactive oxygen species (ROS) generation, mitochondrial membrane potential (MMP) change were examined. The results indicate that a generation dependent cytotoxicity of the PAMAM dendrimers was found to be correlated well with the number of amine group at surface area (G2<G3). Nevertheless, PAMAM dendrimer G2 showed only minor toxicity with low ability to induce apoptosis, low suppress on cell proliferation and small effect on in vitro development. Interestingly, the intracellular ROS level and mitochondrial membrane potential on mouse blastocyst was not altered by PAMAM dendrimer G2. This study provided an important insight that PAMAM dendrimer G2 has low cytotoxicity at the cellular level.
