In this study, the anti-tumor activity effect of a Chinese herb Irkutsk Anemone Rhizome extract (IAR) on human osteosarcoma HOS and U-2OS cells was determined by using cell viability, Western blot and flow cytometry analysis. Cell viability assays revealed that IAR significantly decreased the viability of HOS and U-2OS cells in a dose-dependent manner. With the same IAR dose treatment, cell viability in human osteoblast hFOB1.19 cells was also determined, to identify the most effective and safe dose. Increases in cell shrinkage, Sub-G1-fragments and the activation of caspase-3 and degradation of PARP indicate that IAR-induced cell death may be mediated through the apoptotic signaling pathway in HOS and U-2OS cells. RT-PCR analysis also confirmed up-regulation of PPP1R15A, HSPA1B and DDIT4 by IAR treatment. The findings of anticancer activity in IAR and the identification of some potential target genes raise expectation that IAR may become a useful therapeutic drug for human osteosarcoma.
