We dedicated to find out a compound can effectively inhibit inflammatory progression and less-side effect. The experiment discussed Toddalia asiatica ethyl acetate later extraction and its one of compound, pimpinellin, on LPS-induced RAW264.7 cells. At first, we examined T.asiatica EA layer extraction and pimpinellin by MTT assay to make sure they don’t have cytotoxicity. Then we can keep doing the following experiments. After we synthesized all the results, found out that T.asiatica EA layer extraction and pimpinellin can efficaciously block the production of iNOS and COX-2. Moreover, we discovered both have ability to suppress the expression of NF-kB and MAPK pathway. After we treated T.asiatica EA layer extraction and pimpinellin, they can significantly decrease the content of proinflammatory cytokine. We used pimpinellin to proceed more upstream pathway. Focus on PI3K/Akt pathway, pimpinellin can inhibit the output of PI3K and p-Akt. Pimpinellin also can activate the expression of HO-1 in order to low down the oxidative damage in cells. To summarize above, we drew inference about T.asiatica EA layer extraction and pimpinellin have ability to reduce the production of iNOS, COX-2, TNF-a, IL-1b, IL-6 through PI3K/Akt, NF-kB, MAPK pathway, achieving anti-inflammatory in the end. 黃世勳 2014 學位論文 ; thesis 143 zh-TW
