Summary: | 碩士 === 長庚大學 === 生物醫學研究所 === 102 === G protein coupled receptor 56 (GPR56, also known as TM7XN1) is a member of G protein-coupled receptor (GPCR) superfamily. It has a large N-terminal extracellular domain, which contains a GPCR proteolysis site (GPS) and many potential O‐ and N‐glycosylation sites highly similar to the mucin-like proteins. It has been shown that several ligands can bind to the N-terminal extracellular domain of GPR56. Previous studies have shown that, over-expressing wild type GPR56 in tumor cells, may induce IL-6 secretion. IL-6 is a key factor for mediating tumor progression through activation of STAT-mediated signal transduction and stimulation of cell proliferation and migration. To investigate the relationship between GPR56 and IL-6 secretion, we had used IL-6 blocking antibody for research. We also used V2HS-111640 IL-6 knockdown plasmid for research, but have not succeeded yet.
Moreover, previous studies also showed that cell over-expressing soluble GPR56 may also induce IL-6 secretion. We purify soluble GPR56 for investigating the relationship between soluble GPR56 and cell surface ligands or receptors in melanoma cell. We have shown that purified soluble GPR56 is able to bind ligands of GPR56 and stimulate melanoma migration.
We designed the sandwich ELISA by using purified soluble GPR56 and GPR56 antibodies to measure the concentration of soluble GPR56 in cell conditioned medium or human serum. We found that the concentration of soluble GPR56 in rheumatoid arthritis patient serum is higher than in healthy control. The relevance of soluble GRP56 and diseases rely on
further research in the future.
