Summary:碩士 === 國立中正大學 === 光機電整合工程研究所 === 102 === This work, we demonstrate that the combination of the localized surface plasmonic resonance (LSPR) of silver nanoparticles and strong local filed effect of guided-mode resonance (GMR) in a resonance waveguide grating to produce surface enhanced fluorescence for bio-sensing application. The RWG was fabricated by using holographic lithography and E-gun deposition technique, and then silver nanoparticles were deposits on its surface of the grating by using thermal evaporation method. For biosensing measurement, the Cy3-tagged streptavidin bio-molecules bonded to BSA-bt layer was used as a fluorescent dye, and a 532nm solid-state diode laser was employed as an excitation beam source. The surface enhanced fluorescence can be observed up to 5.8 fold enhancement due to the LSPR of silver nanoparticles. The fluorescence can further promote when the wavelength of the excitation laser resonate with the GMR. Comparing with a flat area, the multiplicative enhancement of fluorescent signal is up to 14.3 fold.
