D) common herbal constituents of healthy tea. The benefits of the healthy tea is associated to prevent liver damage in Taiwan. The study was aimed to determine the antioxidant, anti-inflammatory activities of heat-treated, total phenolic contents and concentration of flavonoids in the four herbs.
In addition, we used human hepatoma cells (Hun-7 and Hep-G2) to test the cytotoxicity of four ethanol extract. The cytotoxic effect of the herabs was better in Hep G2 than Huh 7, except Tithonia Diversifolia ethanol extract. We also tested the cytotoxicity of two ethanol subfractions (P.T and E.S), which passed through silica gel chromatography and successively eluted with different percentage of methanol / hexane.
Antioxidant activities were evaluated by two assays: an in vitro cell free system using a DPPH (2, 2-diphenyl-1-picrylhydrazyl) assay to assess free radical scavenging, and the other cell culture system using preventive effect against peroxidative damage induced by hydrogen peroxide in normal rat liver cell (clone 9). Cell survival was detected by MTT assay and LDH release assay. The flavonoid concentration and phenolic contents were determined using the rutin and galic acid equivalent, respectively. Anti-inflammatory activities of heat-treated (41.5℃, 15 min) in clone 9 were determined by measuring MTT assay and LDH release assay. A dose dependent correlation between antioxidant activities and the total phenolic contents and flavonoid concentration of the plant extracts, respectively, were found. The results were showed the high phenolic and flavonoid content of plant extracts also exhibited significant anti-heat activity with good cell viability. It was speculated that the four herbs owed a rich source of antioxidants and free radical scavenging compounds. The levels of phenolic and flavonoid compounds of the extracts from the herbs were correlated with the antioxidant and anti-inflammatory activities of heat-treated cell in vitro.
