Summary: | 碩士 === 環球科技大學 === 生物技術研究所 === 101 === Herba Vitis is often used as folk herbal medicine in Taiwan to treat chronic nephritis, hepatitis, trauma, bleeding, rheumatism and myalgia. Vitis thunbergii var. taiwaniana (VTT) is an original plant of Vitis Herba. The VTT belongs to Vitaceae family Vitis genus perennial deciduous trailing vine endemic species in Taiwan. It is not only the Formosan original plant, but also a common medicinal herb in Taiwan.
The aim of this study investigated the possible analgesic and anti-inflammatory mechanisms of ethanolic extract of Vitis thunbergii var. taiwaniana (VTTEtOH). In the meantime, HPLC fingerprints was established and the contents of two active ingredients, resveratrol and quercetin were determined, quantitatively. In this study the analgesic effect was evaluated with two animal models including acetic acid-induced writhing response in mice and formalin-induced paw licking time in mice. Furthermore, we have established λ-carrageenan induced mouse paw edema animal model followed by using histopathological analysis and inflammatory mediators determination to evaluate its pharmacological effect and molecular mechanism.
The results showed that VTTEtOH (100, 200 and 400 mg/kg) significantly decreased the acetic acid-induced writhing response, declined licking time in the late phase of formalin test and reduced edema paw volume at 4th to 5th hours after λ-carrageenan injection. We also found that VTTEtOH could suppress inflammatory mediators cyclooxygenase-2 and nitric oxide, and the concentration of inflammatory cytokines (Interleukin-1β, Interleukin-6 and Tumor necrosis factor-α). Beside that, VTTEtOH also reduced the Malondialdehyde level. Histopathological examination showed that VTTEtOH improvedλ-carrageenan induced-inflammation by reducing the paw edema and neutrophil infiltration. The contents of active ingredients, resveratrol and quercetin in VTTEtOH, were 1.61 mg/g and 0.25 mg/g respectively as determined by High Performance Liquid Chromatography.
This study demonstrated the possible mechanisms for the analgesic and anti-inflammatory effects of VTTEtOH, and provided the evidences for the classical treatment of Vitis thunbergii var. taiwaniana in the inflammatory diseases.
