Summary:碩士 === 大同大學 === 生物工程學系(所) === 101 === Haloferax mediterranei, an extreme halophilic archaeon, can accumulate polyhydroxyalkanoate (PHA) as intracellular reserve granules. In the present works, in order to reduce the production cost, fed-batch fermentation was performed with feeding medium contained glucose and yeast extract as carbon and nitrogen sources, respectively. The fed-batch process revealed that OD520nm and PHA production could be 99.7 and 9.3 g/L, respectively. Based on this result, an industrial scale production processes was designed and subsequently analyzed to estimate the production cost of PHA. According to the market share of 1328 kilogram per year of high pure PHA, in about 1.03 years of the payback time could achieve. The selling price of the PHA is estimated as $ 2 per gram in comparison with the high price of ordinary medical biodegradable polymers PLGA(Poly(Lactide-co-Glycolide)) which usually sell above the price $ 7 per gram. Therefore PHA has great potential for the medical biomaterial market.
