Summary:碩士 === 南台科技大學 === 生物科技系 === 101 === Inonotus obliquus (I. obliquus) is a white rot fungus, belonging to the family Hymenochaetaceae in the Basidiomycota, which grows on living trunks of the mature birch, I. obliquus was used as an effective folk medicine fungus in Russia, Eastern Europe, China, and Japan to treat tuberculosis, gastritis, cancer and other diseases. In the study, we used three grains (barley, white wheat, oats) as the solid-state fermentation substrates, and inoculated I. obliquus 84 strain compared to different solid-state substrates, cultivated into days and extracted methods to phenolic content and antioxidant capacity. The results showed that inoculated I. obliquus 84 into three different grain (barley, white wheat, oats) hot water extract (HWE), the phenol content can improve with the increase days, and inoculated 120 days has the highest phenolic, compared with the control group can improve 9.63, 4.84 and 3.32 times, respectively; phenol content per g of HWE were 49.1, 63.4, 37.5 mg GAE, the highest phenol content is white wheat solid-state fermentation substrate.In ABTS radical scavenging capacity performance, barley, white wheat and oat solid-state fermentation substrates inoculated 120 days, the EC50 is 5.13, 3.99 and 7.09 mg/mL, respectively, as shown that the best substrates is white wheat solid-state fermentation substrate, verify the higher phenol contents, the scavenging capacity increase.Compared to reducing power of the three solid-state fermentation substrate, barely, white wheat and oat after inoculated 120 days, the EC50 were 1.19, 0.88 and 1.65 mg/mL, respectively, that showed white wheat solid-state fermentation substrate is the best, and phenol content into a proportional relationship. On OH radical scavenging capacity performance, barley, white wheat and oats solid-state fermentation substrate inoculated 120 days, the EC50 were 4.36, 7.46 and 6.96 mg/mL, as shown that barley solid-state fermentation substrate is the best. I. obliquus 84 different cereals after cultivatefor 120 days in different extracted method (HWE, EtOH extract, Hot water-EtOH extract), barley and white wheat HWE phenol content is the highest, and the oat EtOH extract phenolic content is the highest, the extract of phenolic content per g of extract were 49.1, 63.4, 45.1 mg GAE.In the ABTS radical scavenging capacity, adlay and oat solid-state fermentation substrate display Hot water-EtOH extract will promote the scavenging capacity, and white wheat solid-state fermentation substrate will higher the scavenging capacity after HWE, the EC50 was 4.76, 2.70 and 3.99 mg/mL, respectively, the oat solid state fermentation product antioxidant capacity best. On reducing power and OH radical scavenging capacity, Barley, white wheat and oat solid-state fermentation product with Hot water-EtOH extract will higher the, and the reducing power EC50 were 0.60, 0.79 and 0.59 mg/mL; OH radical scavenging capacity EC50 were 2.91, 3.55 and 3.02 mg/mL, as shown that adlay and oat solid-state fermentation substrate is the best, I. obliquus 84 cultivation three cereal by ethanol extraction can increased antioxidant activity. Therefore I. obliquus 84 solid-state fermentation cultivation has the potential as the antioxidant foods.
