Therefore, the use of two types of stem cells for the treatment of spinal cord injury in rats, try to find a better treatment strategies. The experimental rats were divided into five groups:(1) Sham(laminectomy only);(2) Saline(laminectomy + SCI + normol saline); (3) HUMSCs(laminectomy + SCI + HUMSCs)、(4) HUHSCs(laminectomy + SCI + HUHSCs )、(5) H/M(laminectomy + SCI + HUHSCs/HUMSCs ). In rats T8 -T9 implementation of the intervertebral disc removal surgery (laminectomy) , and artery clip (F=0.5N) clip for 1 minute caused spinal cord injury, until second day after the injured, via jugular vein catheterization performed cell transplantation, then day 7 blood taken for TNF-α and IL-10 detection. Using the BBB locomotors score to assessment hind limb motor function at Day 1, 4, 7 after the injured. The data showed the HUMSCs treatment group has the best hind limb motor function recovery in BBB score. The degree of motor function recovery were : HUMSCs> H / M> HUHSCs> Saline, and in the serum treatment groups HUMSCs can also see a significant increase in IL-10 concentration that play the effect of reducing inflammatory mediators TNF-α. In this experiment confirmed that all spinal cord injury treatment groups, HUMSCs for hindlimb motor function recovery with the best results, and merge two cell types common treatment effect between HUMSCs and HUHSCs. 陳勝咸 102 學位論文 ; thesis 38 zh-TW |
