In Taiwan nervous necrosis virus is the most devastating disease which particularly occurred in grouper’s larvae, where the mortality rate is more than 90%. Conversely, iridovirus has been confirmed that could cause systemic infection in over more than 20 species of fishes. It has high infectivity and high mortality on fishes. Both of these two viruses lead to economic lost in aquaculture industry in Taiwan. Hence, developing an efficient antiviral strategy has become an important issue. Type I interferon system is a rapid and powerful antiviral defense mechanism in vertebrates. When the cell infected by virus interferon well induction of anti-viral molecules, such as Mx protein, and several apoptotic pathways. In this study, we produced grouper type I interferon in Escherichia coli (E. coli) system. Investigating that the type I interfere antiviral function affected in nervous necrosis virus and iridescent virus, as well as discussing cell protective effect treated IFN in different time and different mode. We found that Mx gene expressed after 12 hr when synthetic type I interferon treated cells, further the cytopathic effect (CPE) was delayed increased for survival of cells. Viruses’ replication was also inhibited as shown by real-time RT-PCR assay. In nervous necrosis virus infection, IFN pre- treated can reduce the virus titer in the range of 10 to 30 fold whereas in grouper iridovirus virus infection, IFN pre- treated can reduce the virus titer in the range of 5 to 50 fold.
