There are two pathways for protein degradation in Eukaryote: lysosomal pathway and ubiq-uitin-proteasome system. In previous studies showed that there are high abnormally high levels of 26S proteasome activity in patients with lung cancer, liver cancer or leukemia. Those patients obtain effective cure after treatment of proteasome inhibi-tors. In our previous studies, we found that 26S proteasome activity is inhibited by herbal extracts: methyl gallate (M)、gallic acid (G)、ellagic acid (E) and tannic acid (T). To know the four compounds effected on 26S proteasomal subunits, the interac-tion of Rpt1, 20S core, ubiquitin and ubiquitin-specific processing protease 47 (Usp47) were detected by M, G, E, and T. Glycerol density gradient and ultracentri-fugation were used to separate protein-protein-interaction, and then samples were analyzed by SDS-PAGE and Western blot. In conclusions, the interaction of Ub, Usp47, and Rpt1 of 26S proteasome was effected by M and G. The interaction of Ub, Usp47, and Rpt1 of 26S proteasome was effected by T and E. However the interac-tion of Usp47 and Rpt1 of 19S proteasome was effected by T, M, and G. The inter-action of Ub, Usp47, and Rpt1 of 19S proteasome was effected by E.
