Summary: | 碩士 === 國立高雄師範大學 === 生物科技系 === 101 === Isogenic soil- and mouse-derived strains of B. pseudomallei were made by incubation in the soil and growth in mice, respectively. After incubation in the soil, phenotypic characteristics such as the fatty acid composition, 3-hydroxytetradecanoic acid (an indicator of LPS) , biofilm formation, flagellar expression and ultrastructure on bacterial surface were altered. The varied pathogenesis was determined between two isogenic strains. After infection intravenously, fatal melioidosis resulted from the mouse-derived strains, whereas approximately 70% of the infections with soil-derived strains that resulted in fulminating melioidosis entered remission. In the instances of murine melioidosis due to the soil-derived strains, the degree of bacterial hepatitis and the inflammatory cytokine response gradually decreased after 6 d of infection, and the ability of these strains to invade the bone marrow was significantly attenuated. We suggest that specific phenotypic and pathogenic patterns can be induced by infection with B. pseudomallei cultured in different (soil vs mouse) environments.
